The sim uses 6 bowlers. No more and no less. You will have to mark 6 bowlers.

I am aware of that but my question is will the simulator pick the 6 bowlers who I choose or randomly.

Eg : I have 4 main bowlers and 3 All rounders. I choose one of the all-rounder not to bowl, but is there a chance that the simulator can choose that bowler to be the 6th ?

Yes, sim will definitely select the 6th bowler if it goes for 20 overs and it is random in which order bowlers bowl.

Sim will select only those 6 who you have marked. If you don't mark the 6th bowler then anyone from your lineup will be selected randomly.
